Indications

  • Acute and delayed nausea and vomiting
  • Uncontrolled nausea and vomiting
  • Chemotherapy-induced nausea and vomiting (CINV)
  • Radiotherapy-induced nausea and vomiting (RINV)
  • Post-operative & Post-discharge nausea and vomiting (PONV & PDNV)

Pharmacology

  • 5-HT3 receptor antagonist
  • Strong binding affinity for 5-HT3 receptor
  • Little or no affinity for other receptors
  • Inhibition of serotonin actions
  • Binding affinity to 5-HT3 receptor
  • Linear dose-proportional pharmacokinetics
  • Metabolism via kidney and liver
  • Longer half-life
  • Greater 5-HT3 receptor binding affinity

Dosage & Administration

  • Adult tablet dosage: 0.5 mg daily
  • Adult IV dosage: 0.075 mg over 10 seconds
  • Administered prior to chemotherapy or radiotherapy
  • Children dosage: 20 mcg/kg body weight, maximum 1.5 mg

Interaction

  • Safe administration with corticosteroids, analgesics, antiemetics, antinauseants, antispasmodics, and anticholinergic agents
  • No significant pharmacokinetic interactions with metoclopramide
  • Low potential for clinically significant drug interactions

Contraindications

  • Hypersensitivity to the drug or any of its components

  • Unknown excretion in breast milk
