Indications
- Peptic ulcer diseases
- Gastroesophageal reflux diseases
- Ulcer induced by non-steroidal anti-inflammatory drugs (NSAIDs)
- Eradication of Helicobacter pylori (in combination with antibiotics)
- Zollinger-Ellison Syndrome
Pharmacology
- Proton pump inhibitor
- Suppression of gastric acid production
- Inhibition of basal and stimulated gastric acid secretion
Dosage & Administration
- Dosage for benign gastric ulcer: 40 mg daily in the morning for 4 weeks, maintenance dose is 20 mg daily
- Dosage for gastro-esophageal reflux disease: 20-40 mg daily in the morning for 4 weeks, maintenance dose is 20 mg daily
- Dosage for duodenal ulcer: 40 mg daily in the morning for 2 weeks
- Dosage for Zollinger-Ellison Syndrome: Initially 80 mg once daily, adjusted according to response
- Dosage for IV injection: 40 mg once daily for 7-10 days
- Dosage for prevention of rebleeding in peptic ulcer: IV 80 mg, followed by 8 mg/hour infusion for 72 hours
Interaction
- No significant drug interactions observed in clinical studies
Contraindications
- Known hypersensitivity to any of the components of the formulation
Side Effects
- Well tolerated in both short-term and long-term treatment
- Common side effects: Headache, diarrhea
- Rarely included side effects: Abdominal pain, flatulence, rash, insomnia, hyperglycemia
Pregnancy & Lactation
- US FDA Pregnancy Category: B
- No adequate and well-controlled studies in pregnant women
- Excreted in human milk
- Decision to be made whether to discontinue nursing or discontinue the drug, taking into account the benefit of the drug to the mother
Precautions & Warnings
- Tablet should not be split, chewed or crushed
- Long-term therapy may lead to malabsorption of cyanocobalamin (Vitamin B12)
- May increase the risk of osteoporosis related disorders
Overdose Effects
- No known symptoms of overdosage in humans
- Highly protein bound, not readily dialyzable

Reconstitution
- IV injection is for intravenous administration only
- IV infusion should be given over a period of approximately 15 minutes
- Reconstituted solution may be stored at room temperature for a maximum of 4 hours
