Indications
- Peptic ulcer diseases
- Gastroesophageal reflux diseases
- Ulcer induced by non-steroidal anti-inflammatory drugs (NSAIDs)
- Eradication of Helicobacter pylori (in combination with antibiotics)
- Zollinger-Ellison Syndrome
Pharmacology
Proton pump inhibitor that suppresses the final step in gastric acid production by binding to the H+/K+ATPase enzyme system
Dosage & Administration
- 40 mg daily for benign gastric ulcer
- 20-40 mg daily for gastro-esophageal reflux disease
- 40 mg daily for duodenal ulcer
- 40 mg twice daily with antimicrobial agents for duodenal ulcer associated with Helicobacter pylori
- 20 mg daily for prophylaxis of NSAID-associated gastric or duodenal ulcer
- Initially 80 mg once daily for Zollinger-Ellison Syndrome, adjusted according to response
- 40 mg once daily for duodenal ulcer and gastric ulcer, associated gastroesophageal reflux disease, and prevention of rebleeding in peptic ulcer via IV injection
Interaction
No significant drug interactions observed in clinical studies
Contraindications
Contraindicated in patients with known hypersensitivity to any of the components of the formulation
Side Effects
- Well tolerated in both short-term and long-term treatment
- Common side effects include headache and diarrhea
- Rare side effects include abdominal pain, flatulence, rash, insomnia, and hyperglycemia
Pregnancy & Lactation
US FDA Pregnancy Category B, excreted in human milk, use during pregnancy only if clearly needed
Precautions & Warnings
- Tablet should not be split, chewed, or crushed
- Long-term therapy may lead to malabsorption of cyanocobalamin (Vitamin B12) and increase the risk of osteoporosis related disorders
Overdose Effects
No known symptoms of overdosage in humans, not readily dialyzable

Reconstitution
- IV injection should be given as a slow intravenous injection
- IV infusion should be given over a period of approximately 15 minutes
- Reconstituted solution may be stored at room temperature for a maximum of 4 hours
