Indications
- Neuropathic pain associated with diabetic peripheral neuropathy (DPN)
- Postherpetic neuralgia (PHN)
- Adjunctive therapy for the treatment of partial-onset seizures in patients 1 month of age and older
- Fibromyalgia
- Neuropathic pain associated with spinal cord injury
Pharmacology
Structural derivative of the inhibitory neurotransmitter gamma-aminobutyric acid (GABA) that binds with high affinity to the alpha2-delta site in central nervous system tissues
Dosage
- Maximum recommended dose of 100 mg three times a day for neuropathic pain associated with diabetic peripheral neuropathy
- Recommended dose of 75 to 150 mg two times a day for postherpetic neuralgia in adults
- Recommended dose of 300 to 450 mg/day for management of fibromyalgia
- Recommended dose range of 150 to 600 mg/day for neuropathic pain associated with spinal cord injury
- Dosing instructions for conversion from Pregabalin capsules to Pregabalin CR capsule tablet
Administration
- Route of administration: Oral route, with or without food for Pregabalin, and after an evening meal for Pregabalin CR tablet
- Special administration instructions: Swallow whole, do not split, crush or chew Pregabalin CR tablet. When discontinuing, taper gradually over a minimum of 1 week.
Drug interaction
Unlikely to be involved in significant pharmacokinetic drug interactions
Contraindications
Contraindicated in patients with known hypersensitivity to Pregabalin or any of its components

- Weight gain
- Thinking abnormalities
- Increased weight and increased appetite in pediatric patients
Pregnancy & Lactation
No adequate and well-controlled studies in pregnant women; small amounts detected in the milk of lactating women; breastfeeding not recommended
Precautions & Warnings
- Angioedema may occur and require immediate discontinuation of Pregabalin
- Increased risk of suicidal thoughts or behavior
- Respiratory depression may occur with concomitant CNS depressants
- May cause dizziness and somnolence affecting ability to drive or operate machinery
- Increased seizure frequency or other adverse reactions if rapidly discontinued, should be withdrawn gradually over a minimum of 1 week
- May cause peripheral edema, caution when coadministering with thiazolidinedione antidiabetic agents
Use in Special Populations
Safety and effectiveness not established in pediatric patients for neuropathic pain, postherpetic neuralgia, fibromyalgia and below 1 month of age for partial onset seizures

- Agitation and restlessness
- Seizures
- Heart block
