Description
- Nepco is indicated for neuropathic pain associated with diabetic peripheral neuropathy, postherpetic neuralgia, adjunctive therapy for partial-onset seizures, fibromyalgia, and neuropathic pain associated with spinal cord injury.
- Pregabalin is a structural derivative of the inhibitory neurotransmitter gamma-aminobutyric acid (GABA).
- Dosage varies based on indication and patient's condition, ranging from 75 mg to 660 mg daily.

Dosage
- Maximum recommended dose of Pregabalin is 100 mg three times a day for DPN
- Recommended dose for PHN is 75 to 150 mg two times a day or 50 to 100 mg three times a day
- Recommended dose for fibromyalgia is 300 to 450 mg/day
- Recommended dose range for neuropathic pain associated with spinal cord injury is 150 to 600 mg/day

Contraindications
- Contraindicated in patients with known hypersensitivity to Pregabalin or any of its components
Side Effects
- Common side effects in adults: dizziness, somnolence, dry mouth, edema, blurred vision, weight gain, thinking abnormal
- Common side effects in pediatric patients: increased weight, increased appetite
Pregnancy & Lactation
- No adequate and well-controlled studies in pregnant women
- Small amounts detected in the milk of lactating women
- Breastfeeding not recommended due to potential risk of tumorigenicity
Precautions & Warnings
- Risk of angioedema and respiratory compromise
- Increased risk of suicidal thoughts or behavior
- Respiratory depression may occur when used with concomitant CNS depressants
- May cause dizziness and somnolence, impacting ability to drive or operate machinery
- Caution needed when coadministering with thiazolidinedione antidiabetic agents
Use in Special Populations
- Safety and effectiveness not established in pediatric patients for certain indications
- Safety and effectiveness not established for pediatric patients below the age of 1 month for adjunctive therapy for partial onset seizures

Overdose Effects
- Signs and symptoms include reduced consciousness, depression/anxiety, confusional state, agitation, restlessness, seizures, heart block

- General supportive care indicated including monitoring of vital signs and observation of the clinical status of the patient
