Indications

  • Neuropathic pain associated with diabetic peripheral neuropathy (DPN)
  • Postherpetic neuralgia (PHN)
  • Adjunctive therapy for the treatment of partial-onset seizures in patients 1 month of age and older
  • Fibromyalgia
  • Neuropathic pain associated with spinal cord injury

Pharmacology

  • Structural derivative of the inhibitory neurotransmitter gamma-aminobutyric acid (GABA)
  • Binds with high affinity to the alpha2-delta site in central nervous system tissues

Dosage

  • Maximum recommended dose of 100 mg three times a day for diabetic peripheral neuropathy (DPN)
  • Recommended dose of 75 to 150 mg two times a day or 50 to 100 mg three times a day for postherpetic neuralgia (PHN)
  • Recommended dose of 300 to 450 mg/day for fibromyalgia
  • Recommended dose range of 150 to 600 mg/day for neuropathic pain associated with spinal cord injury
  • Dosage conversion from Pregabalin capsules to Pregabalin CR capsule tablet

Administration

  • Can be taken with or without food
  • Pregabalin CR tablet should be administered after an evening meal
  • Should be swallowed whole and should not be split, crushed or chewed

Drug Interaction

  • Unlikely to be involved in significant pharmacokinetic drug interactions

Contraindications

  • Hypersensitivity to Pregabalin or any of its components

Pregnancy & Lactation

  • Potential risk to fetus, not recommended during breastfeeding

Precautions & Warnings

  • Angioedema can occur and may be associated with life threatening respiratory compromise requiring emergency treatment
  • Increased risk of suicidal thoughts or behavior
  • Respiratory depression may occur when used with concomitant CNS depressants
  • May cause dizziness and somnolence, impairing ability to drive or operate machinery

Use in Special Populations

  • Safety and effectiveness in pediatric patients have not been established
  • Safety and effectiveness in pediatric patients below the age of 1 month have not been established

Overdose Effects

  • Reduced consciousness
  • Depression/anxiety
  • Confusional state
  • Agitation and restlessness
  • Seizures
  • Heart block

  • Primary anti-epileptic drugs
