Indications
- Neuropathic pain associated with diabetic peripheral neuropathy (DPN)
- Postherpetic neuralgia (PHN)
- Adjunctive therapy for the treatment of partial-onset seizures in patients 1 month of age and older
- Fibromyalgia
- Neuropathic pain associated with spinal cord injury
Pharmacology
- Structural derivative of the inhibitory neurotransmitter gamma-aminobutyric acid (GABA)
- Binds with high affinity to the alpha2-delta site in central nervous system tissues
Dosage
- Maximum recommended dose of 100 mg three times a day for diabetic peripheral neuropathy (DPN)
- Recommended dose of 75 to 150 mg two times a day or 50 to 100 mg three times a day for postherpetic neuralgia (PHN)
- Recommended dose of 300 to 450 mg/day for fibromyalgia
- Recommended dose range of 150 to 600 mg/day for neuropathic pain associated with spinal cord injury
- Dosage conversion from Pregabalin capsules to Pregabalin CR capsule tablet
Administration
- Can be taken with or without food
- Pregabalin CR tablet should be administered after an evening meal
- Should be swallowed whole and should not be split, crushed or chewed
Drug Interaction
- Unlikely to be involved in significant pharmacokinetic drug interactions
Contraindications
- Hypersensitivity to Pregabalin or any of its components

- Blurred vision
- Weight gain
- Difficulty with concentration/attention
Pregnancy & Lactation
- Potential risk to fetus, not recommended during breastfeeding
Precautions & Warnings
- Angioedema can occur and may be associated with life threatening respiratory compromise requiring emergency treatment
- Increased risk of suicidal thoughts or behavior
- Respiratory depression may occur when used with concomitant CNS depressants
- May cause dizziness and somnolence, impairing ability to drive or operate machinery
Use in Special Populations
- Safety and effectiveness in pediatric patients have not been established
- Safety and effectiveness in pediatric patients below the age of 1 month have not been established
Overdose Effects
- Reduced consciousness
- Depression/anxiety
- Confusional state
- Agitation and restlessness
- Seizures
- Heart block
