Indications
- Neuropathic pain associated with diabetic peripheral neuropathy (DPN)
- Postherpetic neuralgia (PHN)
- Adjunctive therapy for the treatment of partial-onset seizures in patients 1 month of age and older
- Fibromyalgia
- Neuropathic pain associated with spinal cord injury
Pharmacology
- Structural derivative of the inhibitory neurotransmitter gamma-aminobutyric acid (GABA)
- Binds to the alpha2-delta site in central nervous system tissues
Dosage
- Maximum recommended dose of 100 mg three times a day for neuropathic pain associated with diabetic peripheral neuropathy in adults (DPN)
- Recommended dose of 75 to 150 mg two times a day for postherpetic neuralgia in adults (PHN)
- Recommended dose of 300 to 450 mg/day for management of fibromyalgia in adults
- Recommended dose range of 150 to 600 mg/day for neuropathic pain associated with spinal cord injury in adults
Administration
- Oral route of administration
- Can be taken with or without food
- Pregabalin CR tablet should be administered after an evening meal
- Should be swallowed whole and not be split, crushed or chewed
Drug Interaction
- Unlikely to be involved in significant pharmacokinetic drug interactions
Contraindications
- Contraindicated in patients with known hypersensitivity to Pregabalin or any of its components
Side Effects
- Dizziness
- Somnolence
- Dry mouth
- Edema
- Blurred vision
- Weight gain
- Difficulty with concentration/attention
Pregnancy & Lactation
- No adequate and well-controlled studies with pregabalin in pregnant women
- Potential risk to a fetus
- Breastfeeding is not recommended during treatment with pregabalin
Precautions & Warnings
- Angioedema can occur and may be associated with life threatening respiratory compromise
- Antiepileptic drugs, including Nerfid, increase the risk of suicidal thoughts or behavior
- Respiratory depression may occur with Nerfid when used with concomitant CNS depressants
- Nerfid may cause dizziness and somnolence
- Increased seizure frequency or other adverse reactions may occur if Nerfid is rapidly discontinued
- Caution should be exercised when coadministering Nerfid and thiazolidinedione antidiabetic agents
Use in Special Populations
- Safety and effectiveness in pediatric patients have not been established for various conditions
- Safety and effectiveness of Nerfid extended-release tablet in pediatric patients have not been established
Overdose Effects
- Signs and symptoms include reduced consciousness, depression/anxiety, confusional state, agitation, restlessness, seizures, and heart block
- No specific antidote
