Indications
- Control severe nausea and vomiting caused by radiation therapy, cancer chemotherapy, surgery
- Relieve nausea, vomiting, dizziness, vertigo associated with Meniere's disease
- Treatment of psychotic illness such as schizophrenia
- Short-term treatment of generalized non-psychotic anxiety
Pharmacology
Dopamine & histamine antagonist; blocks histamine H & dopamine D2 neurotransmitter receptors
Dosage & Administration
- Antiemetic:
- Dosage for children based on weight
- Adult dosage: 5-10 mg 3-4 times/day
- Antipsychotic:
- Dosage for children based on age
- Adult dosage: 5-10 mg 3-4 times/day; up to 150 mg/day in severe cases
- Nonpsychotic anxiety:
- Adult dosage: 15-20 mg/day in divided doses
- Elderly dosage: Initial 2.5-5 mg 1-2 times/day; increase gradually
- Administration: May be administered without regard to the meal
Interaction
- Alcohol, barbiturate & sedatives may increase CNS depressant action
- Interference with absorption of other drugs
- Interference with plasma concentration of Propanolol and Phenobarbital
Contraindications
- Hypersensitivity to prochlorperazine or any component
- Severe CNS depression, coma
- Not recommended for children <2 years of age or <10 kg
Side Effects
- Drowsiness
- Muscle spasms
- Fine worm-like tongue movements
- Rhythmic face, mouth, or jaw movements
- Skin rash
- Yellowing of skin or eyes
Pregnancy & Lactation
No reported adverse effects during pregnancy & lactation
Precautions & Warnings
- Caution in tasks requiring alertness
- Extreme drowsiness with alcohol use
- Increased sensitivity to sunlight
- Reduced sweating, more susceptible to heatstroke
Overdose Effects
Deep sleep, coma, abnormal involuntary muscle movements, hypotension
