Indications
- Treatment of active duodenal ulcer
- Treatment of active benign gastric ulcer
- Symptomatic erosive or ulcerative gastro-esophageal reflux disease (GERD)
- Long-term management of gastro-esophageal reflux disease (GERD maintenance)
- Symptomatic treatment of moderate to very severe gastro-esophageal reflux disease (symptomatic GERD)
- Treatment of Zollinger-Ellison Syndrome
- Eradication of Helicobacter pylori in patients with peptic ulcer disease
Pharmacology
- Suppression of gastric acid secretion
- Inhibition of gastric H+/K+-ATPase
Dosage
- Recommended dose for active duodenal ulcer and active benign gastric ulcer is 20 mg once daily in the morning
- Recommended oral dose for erosive or ulcerative gastro-esophageal reflux disease (GERD) is 20 mg once daily for four to eight weeks
- For long-term management of GERD, a maintenance dose of rabeprazole sodium 20 mg or 10 mg once daily can be used
- 10 mg once daily for symptomatic treatment of moderate to very severe gastro-esophageal reflux disease (symptomatic GERD)
- Treatment of GERD in pediatric patients 1 to 11 years of age: 5 mg or 10 mg once daily depending on weight
- Recommended starting dose for Zollinger-Ellison Syndrome is 60 mg once a day, titrated upwards as necessary
- Combination therapy for H. pylori eradication: rabeprazole sodium 20 mg twice daily
Administration
- Tablets should be taken in the morning, before eating
- Tablets should not be chewed or crushed, but swallowed whole
Interaction
- Potential interaction with pH-dependent compounds
- Co-administration with ketoconazole or itraconazole may decrease antifungal plasma levels
- No interaction with liquid antacids observed
- Not recommended to be co-administered with atazanavir
Contraindications
- Hypersensitivity to the active substance or excipients
- Contraindicated in pregnancy and during breastfeeding

- Possible increased risk of hip, wrist, and spine fractures with long-term use, especially in the elderly
- Risk of cross-hypersensitivity reactions with other proton pump inhibitors or substituted benzimidazoles
- Post marketing reports of blood dyscrasias and hepatic enzyme abnormalities
- Monitoring of magnesium levels for patients on prolonged treatment
Use in Special Populations
- No dosage adjustment necessary for renal or hepatic impairment
- Not recommended for use in children due to lack of data
Overdose Effects
- Maximum established exposure not exceeding 60 mg twice daily or 160 mg once daily
- Effects generally minimal, reversible without further medical intervention
