Indications

Pharmacology
Rabeprazole suppresses gastric acid secretion by inhibiting the gastric H+/K+-ATPase at the secretory surface of the gastric parietal cell.
Dosage
- Active Duodenal Ulcer and Active Benign Gastric Ulcer: 20 mg once daily in the morning
- Erosive or Ulcerative Gastro-Esophageal Reflux Disease (GERD): 20 mg once daily for four to eight weeks
- Gastro-Esophageal Reflux Disease Long-term Management (GERD Maintenance):
- 20 mg once daily
- 10 mg once daily based on patient response
- Symptomatic treatment of moderate to very severe Gastro-Esophageal Reflux Disease (symptomatic GERD):
- 10 mg once daily
- 10 mg once daily on-demand when needed
- Treatment of GERD in pediatric patients 1 to 11 years of age (Less than 15 kg): 5 mg once daily for 12 weeks
- Treatment of GERD in pediatric patients 1 to 11 years of age (15 kg or more): 10 mg once daily for 12 weeks
- Zollinger-Ellison Syndrome:
- 60 mg once daily
- titrated upwards to 120 mg/day based on individual patient needs
- Eradication of H. pylori: Rabeprazole sodium 20 mg twice daily, clarithromycin 500 mg twice daily and amoxicillin 1g twice daily for 7 days
Administration
Should be taken in the morning, before eating, and should not be chewed or crushed, but swallowed whole.
Interactions
- Positive Interaction: No interaction with liquid antacids was observed.
- Negative Interaction: Co-administration with ketoconazole or itraconazole may result in a significant decrease in antifungal plasma levels.
Contraindications
- Hypersensitivity to the active substance or to any of the excipients
- Contra-indicated in pregnancy and during breastfeeding

- Cross-hypersensitivity reactions with other proton pump inhibitors or substituted benzimidazoles cannot be excluded
- Hypomagnesaemia and its symptoms should be monitored in long-term users
- May reduce the absorption of vitamin B12
Use in Special Populations
- No dosage adjustment is necessary for patients with renal or hepatic impairment
- Not recommended for use in children due to a lack of data on safety and efficacy
Overdose Effects
- Maximum Established Exposure: 60 mg twice daily, or 160 mg once daily
- Effects: Generally minimal, representative of the known adverse event profile and reversible without further medical intervention
- Antidote: No specific antidote known. Symptomatic and general supportive measures should be utilized.
