Description
- Profast Gastro-resistant tablets are indicated for the treatment of active duodenal ulcer, active benign gastric ulcer, symptomatic erosive or ulcerative gastro-esophageal reflux disease (GERD), and more.
- Rabeprazole suppresses gastric acid secretion by inhibiting the gastric H+/K+-ATPase at the secretory surface of the gastric parietal cell.
Indications
- Treatment of active duodenal ulcer
- Treatment of active benign gastric ulcer
- Symptomatic erosive or ulcerative gastro-esophageal reflux disease (GERD)
- Gastro-esophageal Reflux Disease Long-term Management (GERD Maintenance)
- Symptomatic treatment of moderate to very severe gastro-esophageal reflux disease (symptomatic GERD)
- Zollinger-Ellison Syndrome
- Eradication of Helicobacter pylori in patients with peptic ulcer disease
Dosage
- 20 mg to be taken once daily in the morning for active duodenal ulcer and active benign gastric ulcer
- 20 mg to be taken once daily for four to eight weeks for erosive or ulcerative GERD
- Rabeprazole sodium 20 mg or 10 mg once daily for GERD maintenance
- 10 mg once daily for symptomatic treatment of moderate to very severe GERD
- 60 mg once a day for Zollinger-Ellison Syndrome
- Combination therapy for H. pylori eradication: Rabeprazole sodium 20 mg twice daily, clarithromycin 500 mg twice daily, and amoxicillin 1g twice daily for 7 days
Administration
- Rabeprazole tablets should be taken in the morning, before eating, and swallowed whole
Interaction
- Interaction with compounds whose absorption is pH dependent may occur
- Co-administration of Profast with ketoconazole or itraconazole may result in a significant decrease in antifungal plasma levels
- No interaction with liquid antacids observed
- PPIs, including rabeprazole, should not be co-administered with atazanavir
Contraindications
- Hypersensitivity to the active substance or excipients
- Contra-indicated in pregnancy and during breastfeeding

Pregnancy & Lactation

Precautions & Warnings
- Regular surveillance for patients on long-term treatment
- Possible increase in the risk of hip, wrist, and spine fracture with long-term high dose use
- Risk of cross-hypersensitivity reactions with other proton pump inhibitor or substituted benzimidazoles
- Caution against chewing or crushing tablets
- Reports of blood dyscrasias and hepatic enzyme abnormalities
Use in Special Populations
- No dosage adjustment is necessary for patients with renal or hepatic impairment
- Not recommended for use in children
Overdose Effects
- Maximum established exposure has not exceeded 60 mg twice daily
- Effects are generally minimal and reversible without further medical intervention
- No specific antidote is known
