Pharmacology

  • Suppression of Gastric Acid Secretion

  • Characterized as a Gastric Proton-Pump Inhibitor

Dosage

  • Recommended Oral Dose for Active Duodenal Ulcer and Active Benign Gastric Ulcer: 20 mg once daily in the morning
  • Recommended Oral Dose for Erosive or Ulcerative Gastro-Esophageal Reflux Disease (GERD): 20 mg once daily for 4-8 weeks
  • For Long-term Management: Maintenance dose of 20 mg or 10 mg once daily

Administration

  • Tablets should be taken in the morning, before eating
  • Tablets should not be chewed or crushed, but swallowed whole

Interaction

  • Potential interaction with pH-dependent compounds like ketoconazole or itraconazole
  • No interaction with liquid antacids observed
  • Atazanavir absorption is pH-dependent, should not be co-administered with PPIs

Contraindications

  • Hypersensitivity to the active substance or excipients
  • Contraindicated in pregnancy and during breastfeeding

  • Potential interference with vitamin B12 absorption

Use in Special Populations

  • No dosage adjustment for patients with renal or hepatic impairment
  • Not recommended for use in children

Overdose Effects

  • Effects are generally minimal and reversible without medical intervention
  • No specific antidote known, treatment should be symptomatic
