Indications

  • Symptomatic treatment of Seasonal & Perennial Allergic Rhinitis and Urticaria

Pharmacology

  • Rupatadine is a long-acting, non-sedative antagonist of histamine H1-receptors
  • Antagonizes the platelet activating factor (PAF)
  • Inhibits degranulation of mast cells
  • Inhibition of the release of cytokines

Dosage & Administration

  • Recommended dose for adults and adolescents is 10 mg once a day
  • Recommended doses for children aged 2 to 11 years based on weight
  • Can be taken with or without food

Interaction

  • Concomitant administration with ketoconazole or erythromycin increases systemic exposure
  • Caution with statins, CNS depressants, alcohol, and inhibitors of isozyme CYP3A4
  • Grapefruit and Grapefruit juice should not be taken simultaneously with Rupatadine

Contraindications

  • Hypersensitivity to Rupatadine or any of the excipients

Side Effects

  • Common: Asthenia, dizziness, drowsiness
  • Uncommon: Appetite increased, arthralgia, back pain, concentration impaired, constipation, cough, diarrhea, dry throat, epistaxis
  • Rare: Palpitations, tachycardia

Pregnancy & Lactation

  • Not recommended during pregnancy unless potential benefit outweighs potential risk for the infant
  • Not recommended during lactation unless potential benefits justify potential risk to the infant

Precautions & Warnings

  • Use with caution in patients with known prolongation of QT interval, uncorrected hypokalemia, and ongoing proarrhythmic conditions
  • Caution in elderly patients
  • Not recommended for patients with impaired kidney or liver function

Use in Special Populations

  • Use with caution in elderly
  • Not established in patients less than 12 years of age
  • Not recommended in patients with renal or hepatic insufficiency

Overdose Effects

  • Common adverse reaction is somnolence
  • Symptomatic treatment and supportive measures for accidental ingestion of very high doses
