Indications
- Symptomatic treatment of Seasonal & Perennial Allergic Rhinitis and Urticaria
Pharmacology
- Rupatadine is a long-acting, non-sedative antagonist of histamine H1-receptors
- Antagonizes the platelet activating factor (PAF)
- Inhibits degranulation of mast cells
- Inhibition of the release of cytokines
Dosage & Administration
- Recommended dose for adults and adolescents is 10 mg once a day
- Recommended doses for children aged 2 to 11 years based on weight
- Can be taken with or without food
Interaction
- Concomitant administration with ketoconazole or erythromycin increases systemic exposure
- Caution with statins, CNS depressants, alcohol, and inhibitors of isozyme CYP3A4
- Grapefruit and Grapefruit juice should not be taken simultaneously with Rupatadine
Contraindications
- Hypersensitivity to Rupatadine or any of the excipients
Side Effects
- Common: Asthenia, dizziness, drowsiness
- Uncommon: Appetite increased, arthralgia, back pain, concentration impaired, constipation, cough, diarrhea, dry throat, epistaxis
- Rare: Palpitations, tachycardia
Pregnancy & Lactation
- Not recommended during pregnancy unless potential benefit outweighs potential risk for the infant
- Not recommended during lactation unless potential benefits justify potential risk to the infant
Precautions & Warnings
- Use with caution in patients with known prolongation of QT interval, uncorrected hypokalemia, and ongoing proarrhythmic conditions
- Caution in elderly patients
- Not recommended for patients with impaired kidney or liver function
Use in Special Populations
- Use with caution in elderly
- Not established in patients less than 12 years of age
- Not recommended in patients with renal or hepatic insufficiency
Overdose Effects
- Common adverse reaction is somnolence
- Symptomatic treatment and supportive measures for accidental ingestion of very high doses
