Indications
- Primary hypercholesterolemia (type IIa and IIb)
- Coronary heart disease
- Elevated plasma cholesterol level
Pharmacology
- Cholesterol lowering agent
- Inhibition of HMG-CoA reductase activity
- Increased LDL receptor density
- Increased removal of LDL cholesterol
- Reduced formation of LDL from VLDL
- Extensively metabolized in the liver
Dosage & Administration
- Usual starting dose: 10 mg/day
- Adjustment of dosage at intervals of not less than four weeks
- Maximum dose: 40 mg daily
- Consideration for dose reduction if cholesterol levels fall below certain thresholds
- No significant renal excretion
- Safety and effectiveness in children and adolescents not established
Interaction
- Concomitant administration with Digoxin
- Enhancement of anticoagulant effect of Warfarin by Coumarin derivatives
- No clinically significant adverse interactions with ACE inhibitors, beta-blockers, calcium channel blockers, diuretics, and NSAIDs
Contraindications
- Active liver disease
- Pregnant and breastfeeding women
- Women of childbearing age without adequate contraception
- Hypersensitivity to any component of the preparation
- Patients with homozygous familial hypercholesterolemia

Precautions & Warnings
- History of liver disease
- High alcohol intake
- Liver function test before and during treatment
- Treatment discontinuation if serum transaminase rises three times the upper limit of normal
- Avoid pregnancy during and for one month after treatment
