Indications

  • Adjunct to diet and exercise

Pharmacology

  • DPP-4 inhibitor
  • Slows inactivation of incretin hormones
  • Increases and prolongs action of hormones
  • Regulates glucose homeostasis
  • Increases insulin synthesis and release
  • Lowers glucagon secretion
  • Selective for DPP-4

Dosage & Administration

  • Recommended dose: 50mg twice a day
  • Recommended dose: 100mg once daily
  • Can be taken with or without food

Interaction

  • Does not alter pharmacokinetics of specific drugs
  • Slightly increases Digoxin concentration

Contraindications

  • History of serious hypersensitivity reaction to sitagliptin

Side Effects

  • Common adverse reactions: headache, upper respiratory tract infection, nasopharyngitis
  • Hypoglycemia risk in combination with sulfonylurea or insulin

  • Pregnancy Category B
  • Safety in lactation not determined

  • Discontinue promptly if pancreatitis suspected
  • Dosage adjustment for renal insufficiency
