Indications
- Erectile Dysfunction (ED)
- Benign Prostatic Hyperplasia (BPH)
- Both Erectile Dysfunction and signs and symptoms of Benign Prostatic Hyperplasia
Pharmacology
- Selective phosphodiesterase type 5 (PDE5) inhibitor
- Increases cGMP in smooth muscle cells
- Causes smooth muscle relaxation
- Increases blood flow into the corpus cavernosum
- Present in smooth muscles of the prostate and bladder wall
- Improves blood flow to the urinary tract
- Widens the opening of the bladder neck
- Results in improved voiding
Dosage & Administration
- Recommended starting dose for Erectile Dysfunction: 10 mg
- Maximum dosing frequency: once daily
- Effective for up to 36 hours
- Recommended dose for Benign prostatic hyperplasia: 5 mg
- Recommended dose for Combined Erectile Dysfunction and Benign prostatic hyperplasia: 5 mg
Interaction
- May interact with Nitrates, Alpha adrenergic blockers, Antihypertensives, Alcohol, Antacids, Ketoconazole, Ritonavir, Erythromycin, Itraconazole, Grapefruit juice, Rifampin, Carbamazepine, Phenytoin, Phenobarbital
Contraindications
- Use of Nitrates
- Hypersensitivity reactions to Tadalafil
Side Effects
- Common side effects: Headache, Dyspepsia, Back pain, Myalgia, Nasal pharyngitis, Nasal congestion
- Rare side effects: Change in Color Vision, Sudden vision loss, Hearing loss, Stevens-Johnson Syndrome, Exfoliative dermatitis, Angina, Stroke, Myocardial infarction, Severe hypotension, Tachycardia
Pregnancy & Lactation
- Tadalafil assigned to pregnancy category B by the USFDA
- Not recommended for use during pregnancy unless benefit outweighs risk
- Caution should be used when administering tadalafil to nursing women
