Indications
- Treats thiamine deficiency such as Beriberi and Wernick's encephalopathy
- Neuritis associated with pregnancy
- Supplementary prophylactic treatment for low dietary intake or impaired absorption of thiamine
Pharmacology
- Thiamine pyrophosphate is the coenzyme for decarboxylation of α-ketoglutaric acid
- Necessary for optimal growth of infants and children
- Not stored in the body and is regularly lost during deficiency
- Deficiency leads to fatigue, anorexia, gastrointestinal disturbance, and neurological symptoms
Dosage administration
- Prophylaxis: 3 to 10 mg daily
- Mild chronic deficiency: 10 to 25 mg daily
- Severe deficiency: 200 to 300 mg daily
Interaction
- No hazardous drug interactions reported
- Acts synergistically with other B-complex vitamins
Contraindications
- Risk of anaphylaxis with repeated parenteral administration
- Mild allergic reactions may lead to anaphylactic shock
- Avoid intravenous administration except for comatose patients
Side effects
- No adverse effects when given orally
- Anaphylactic reactions may occur with large intravenous doses
- Transient mild soreness at intramuscular site
