Description
Used to reduce cholesterol saturation of bile and promote dissolution of gallstones.
Mode of Action
- Reduces cholesterol absorption
- Suppresses liver cholesterol synthesis
- Promotes formation of liquid cholesterol crystal complexes
- Protects cholangiocytes against cytotoxicity
- Stimulates hepatobilliary secretion
- Protects hepatocytes against bile acid-induced apoptosis
Pharmacology
- Absorption: 60-80%
- Peak serum concentration: 30-150 minutes
- Excretion: Small amounts in urine
- Half life: 3.5-5.8 days
Dosage & Administration
- Dissolution of Gallstones: 8-12 mg/kg/day
- Primary Billiary Cirrhosis: 10-15 mg/kg/day
- Acute Viral Hepatitis: 600 mg/day
- Alcoholic Fatty Liver: 300 mg/day
- Primary Sclerosing Cholangitis: 25-30 mg/kg/day
Interaction
Avoid concomitant use with bile-acid binding drugs
Contraindications
- Non-functioning gall-bladder
- Calcified and pigmented gallstones
- Inflammatory bowel disease
