Description
UDCA (Ursodeoxycholic Acid) is used to treat various hepatobiliary disorders. It reduces the cholesterol saturation of bile and promotes the dissolution of gallstones through several mechanisms.
Indications
- Cholestasis (Jaundice)
- Viral Hepatitis
- Alcoholic Fatty Liver
- Primary Billiary Cirrhosis (PBC)
- Primary Sclerosing Cholangitis (PSC)
- Dissolution of Gallstones
- Non-Alcoholic Steato Hepatitis (NASH)
Mode of action
- Decreases secretion of cholesterol in bile
- Reduces cholesterol absorption
- Suppresses liver cholesterol synthesis
- Alters bile composition from supersaturated to unsaturated
- Promotes formation of liquid cholesterol crystal complexes
Dosage and administration
- Dissolution of gallstones: 8-12 mg/kg/day either as single night time dose or in divided doses
- Primary billiary cirrhosis: 10-15 mg/kg/day in 2-4 divided doses
- Acute viral hepatitis: 600 mg/day
- Alcoholic fatty liver: 300 mg/day
- Primary sclerosing cholangitis: 25-30 mg/kg/day
- Nash and gallstone dissolution: 13-15 mg/kg/day
Interaction
UDCA should not be used with drugs that increase bile cholesterol, such as oestrogenic hormones. Concomitant administration with bile-acid binding drugs should be avoided.
Contraindications
- Non-functioning gall-bladder
- Calcified and pigmented gallstones
- Inflammatory bowel disease
