Description
- Reduction of cholesterol saturation of bile
- Promotion of dissolution of gallstones
- Reduction of cholesterol secretion into bile
- Increase in bile acid secretion rate
Pharmacology
- Decrease in secretion of cholesterol in bile
- Reduction of cholesterol absorption
- Suppression of liver cholesterol synthesis
- Altering bile composition from supersaturated to unsaturated
- Promoting formation of liquid cholesterol crystal complexes
- Enhancing removal of cholesterol from the gallbladder into the intestine
- Improvement of cholestatic liver diseases
- Protecting cholangiocytes against cytotoxicity of hydrophobic bile acids
- Stimulating hepatobilliary secretion
- Protecting hepatocytes against bile acid-induced apoptosis
- Complete absorption in the upper intestine
- Variation in time to peak serum concentration (30 to 150 minutes)
Dosage & Administration
- Dissolution of Gallstones: 8-12 mg/kg/day
- Primary Billiary Cirrhosis: 10-15 mg/kg/day
- Acute Viral Hepatitis: 600 mg/day
- Alcoholic Fatty Liver: 300 mg/day
- Primary Sclerosing Cholangitis: 25-30 mg/kg/day
Interaction
- Incompatibility with oestrogenic hormones
- Avoid concomitant administration with bile-acid binding drugs (antacids, charcoal, cholestyramine)
Contraindications
- Non-functioning gall-bladder
- Calcified and pigmented gallstones
- Inflammatory bowel disease
Side Effects

- Gallstone opacilication

Pregnancy & Lactation
- Pregnancy category B
- No evidence of harm reported in pregnancy
- Effective treatment for cholestasis of pregnancy during the last trimester
- No documented problems in humans regarding breastfeeding
