Description

  • Reduction of cholesterol saturation of bile
  • Promotion of dissolution of gallstones
  • Reduction of cholesterol secretion into bile
  • Increase in bile acid secretion rate

Pharmacology

  • Decrease in secretion of cholesterol in bile
  • Reduction of cholesterol absorption
  • Suppression of liver cholesterol synthesis
  • Altering bile composition from supersaturated to unsaturated
  • Promoting formation of liquid cholesterol crystal complexes
  • Enhancing removal of cholesterol from the gallbladder into the intestine
  • Improvement of cholestatic liver diseases
  • Protecting cholangiocytes against cytotoxicity of hydrophobic bile acids
  • Stimulating hepatobilliary secretion
  • Protecting hepatocytes against bile acid-induced apoptosis
  • Complete absorption in the upper intestine
  • Variation in time to peak serum concentration (30 to 150 minutes)

Dosage & Administration

  • Dissolution of Gallstones: 8-12 mg/kg/day
  • Primary Billiary Cirrhosis: 10-15 mg/kg/day
  • Acute Viral Hepatitis: 600 mg/day
  • Alcoholic Fatty Liver: 300 mg/day
  • Primary Sclerosing Cholangitis: 25-30 mg/kg/day

Interaction

  • Incompatibility with oestrogenic hormones
  • Avoid concomitant administration with bile-acid binding drugs (antacids, charcoal, cholestyramine)

Contraindications

  • Non-functioning gall-bladder
  • Calcified and pigmented gallstones
  • Inflammatory bowel disease

Pregnancy & Lactation

  • Pregnancy category B
  • No evidence of harm reported in pregnancy
  • Effective treatment for cholestasis of pregnancy during the last trimester
