Description
- Benzimidazole anthelmintic
- Active against most nematodes and some cestodes
- Treatment of intestinal nematode infections
- Treatment of hydatid disease
- Vermicidal activity
- Ovicidal activity
- Larvicidal activity
- Inhibitory effect on tubulin polymerization
Pharmacology
- Broad spectrum anthelmintic
- Block glucose uptake in susceptible helminths
- Depletion of energy level in helminths
- Immobilization of parasites
- Binding and inhibition of parasite tubulin polymerization
- Metabolized in the liver
- Poorly absorbed from gastrointestinal tract
- Excretion in urine
Dosage & Administration
- 400 mg single dose for specific infections in adults & children over 2 years
- 400 mg daily for 3 days in cases of strongyloidiasis or taeniasis
- 200 mg single dose for children of 1-2 years
- Not recommended for children under 1 year
- Dose varies based on weight for hydatid disease
- 400 mg once daily for 5 days in giardiasis

Contraindications
- Not normally used in neonates
- Dose reduction may be needed in children under 10 kg
- Not recommended during pregnancy or for women thought to be pregnant
- No evidence to suggest altering dose in renal, hepatic, or cardiac failure

- Changes in liver enzymes
- Reversible alopecia (rare)

- Fever
- Blood disorders including leucopenia and pancytopenia
- Allergic shock if cyst leakage
- Convulsion and meningism in cerebral disease

- Category C in US FDA pregnancy rating
- Avoid in pregnancy and lactation unless benefits outweigh risks
Precautions & Warnings
- Blood counts and liver function tests before and during treatment
- Caution in breastfeeding
- Exclude pregnancy before treatment
- Constant medical supervision for Echinococcosis treatment
