Indications
- Reduction of total cholesterol and LDL cholesterol in familial hypercholesterolemia
- Treatment of mixed dyslipidemia (Fredrickson Type Ia and Ib)
- For the treatment of hypertriglyceridemia (Fredrickson Type IV)
- Treatment of dysbetalipoproteinaemia (Fredrickson Type III)
- Reduction of cardiac ischaemic events in patients with coronary artery disease
- Reduction of cholesterol levels in patients with diabetes mellitus or renal transplantation
Pharmacology
- Selective inhibitor of HMG-CoA reductase
- Lowers plasma cholesterol and lipoprotein levels
- Rapid absorption after oral administration
- Extensively metabolized to active derivatives
- Primarily eliminated in bile
- Mean plasma elimination half-life of approximately 14 hours
DosageAdministration
- Initial dose of 10 mg once daily for adults
- Increased to max. 80 mg once daily if necessary
- Dosage range of 10 to 80 mg once daily
- Dosage adjustment based on lipid levels
Interaction
- Increased risk of myopathy with concurrent use of certain drugs
- Altered plasma concentrations with antacid, colestipol, digoxin, erythromycin, oral contraceptives
- No clinically significant effect on prothrombin time when coadministered with warfarin
Contraindications
- Hypersensitivity to any component of the medication
- Active liver disease or unexplained persistent elevations of serum transaminases
- History of serious adverse reaction to prior administration of HMG-CoA reductase inhibitors

- No clinically significant difference in LDL-C reduction between genders
- Renal disease has no influence on plasma concentrations or LDL-C reduction
- Markedly increased plasma concentrations in chronic alcoholic liver disease
