Pharmacology
- Selective inhibitor of HMG-CoA reductase
- Lowering plasma cholesterol and lipoprotein levels
- Inhibiting cholesterol synthesis
- Increasing hepatic LDL receptors
- Rapid absorption after oral administration
- Extensive metabolism by cytochrome P450 3A4
- Elimination primarily in bile
- Plasma elimination half-life of approximately 14 hours
Dosage & Administration
- Adults dosage of 10-80 mg once daily
- Child (10-18 years) dosage of 10-20 mg once daily
- Starting dose of 10 or 20 mg once daily
- Dosage range of 10 to 80 mg once daily
- Lipid level analysis within 2 to 4 weeks of initiation
Interaction
- Increased risk of myopathy with concurrent administration of certain drugs
- Altered plasma concentrations when coadministered with antacid, colestipol, digoxin, erythromycin, oral contraceptives
- No clinically significant effect on prothrombin time when administered with warfarin
Contraindications
- Hypersensitivity to any component
- Active liver disease or unexplained persistent elevations of serum transaminases
- History of serious adverse reaction to prior administration of HMG-CoA reductase inhibitors

Use in Special Populations
- Higher plasma concentrations in elderly subjects
- No clinically significant difference in LDL-C reduction between men and women
- No influence on plasma concentrations or LDL-C reduction in renal disease
