Description
Alfradin DS is a semisynthetic broad-spectrum bactericidal antibiotic, active against infections caused by both gram-positive and gram-negative microorganisms.
Indications
- Treatment of infections caused by sensitive Gram-positive and Gram-negative bacteria
- Upper respiratory tract infections: sinusitis, pharyngitis, tonsillitis, laryngo-tracheo bronchitis, otitis media
- Lower respiratory tract infections: bronchitis, lobar pneumonia, bronchopneumonia
- Urinary tract infections: cystitis, urethritis, pyelonephritis
- Skin and soft tissue infections: abscess, cellulitis, furunculosis, impetigo
Pharmacology
Cephradine inhibits cross-linking process of peptidoglycan in the cell wall of bacteria, leading to lysis of bacteria due to external osmotic pressure
DosageAndAdministration
- OralAdministration:
- Adults:
- Urinary tract infections: 500mg four times daily or 1g twice daily
- Respiratory tract infections: 250-500mg four times daily or 500mg-1g twice daily
- Skin and soft tissue infections: 250-500mg four times daily or 500mg-1g twice daily
- Children:
- Total daily dose: 25-50mg/kg given in two or four equally divided doses
- Otitis media: Total daily dose: 75-100mg/kg given in divided doses
- Elderly: Normal adult dose is appropriate. Patients with impaired renal or hepatic function should be monitored during treatment
- Adults:
- InjectableAdministration:
- Adult: Usual dose: 2-4 gm daily in four equally divided doses, up to 8 gm daily
Interactions
Concomitant use of nephrotoxic drugs may increase risk of kidney damage, diuretics and probenecid enhance renal toxicity

- Prolonged use may result in superinfection
- Care with penicillin hypersensitivity
- May cause false positive urine glucose result
- Dosage adjustment necessary in renal impairment
- Contains lactose, caution in patients with galactose intolerance
UseInSpecialPopulations
- RenalImpairment:
- CrCl>20 ml/min: 500mg every 6 hours
- CrCl: 5-20 ml/min: 250mg every 6 hours
- CrCl: <5 ml/min: 250mg every 50-70 hours
- Chronic, intermittent haemodialysis: 250mg at the start of haemodialysis

StorageConditions
Suspension should be used within 7 days if kept at room temperature or within 14 days if kept in a refrigerator, Injection solutions should be used within 2 hours when kept at room temperature
