Description
- Selective alpha-1 adrenoreceptor competitive blocking agent
- Quinazoline derivative
- Rapid absorption
- Activity within 15 minutes after oral ingestion
Mode of Action
- Selective for alpha-1-adrenoceptors
- Relaxation of smooth muscle in blood vessels and prostate
- Lowering blood pressure
- Improving urinary flow
- Reduction of pressure on the urethra
- Inhibition of catecholamines-induced factors responsible for mitogenesis
- Induction of apoptosis of prostate cells
- Enhancement of expression of transforming growth factor beta-1 (TGF-beta1)
- Upregulation of p27kip1 and the caspase cascade
Dosage & Administration
- Initial dose: 1 mg at bedtime for all patients
- Subsequent dose: Slowly increase to achieve the desired response
- Recommended dose range for BPH: 5-10 mg administered once a day
- Recommended dose range for hypertension: 2-10 mg administered once a day
Interactions
- Possible interaction with ACE inhibitors or diuretics leading to dizziness or related side effects
- No interaction with analgesics/anti-inflammatory, cardiac glycosides, hypoglycemic, antiarrhythmic, anxiolytics/sedatives, antibacterial, hormones/steroids, and drugs used for gout
Contraindications
- Hypersensitivity to Terazosin or its analogues

- Postural hypotension
- Dizziness
- Lack of energy
- Peripheral edema
- Urinary frequency
- Priapism
Pregnancy & Lactation
- Not recommended during pregnancy unless potential benefit justifies potential risk to mother and fetus
- Exercise caution when administered to a nursing mother
Precautions & Warnings
- First dose may cause hypotension
- Caution when administered with other antihypertensive agents
- Avoid driving and hazardous tasks due to possibility of syncope after initiation of therapy
Overdose Effects
- Acute hypotension
- Cardiovascular support is of first importance
- Supine position recommended
- Expansion of plasma volume and noradrenergic vasopressor may be needed
