Description

• Selective alpha-1 adrenoreceptor competitive blocking agent

Indications

• Relief from signs and symptoms of benign prostatic hyperplasia (BPH)
• Treatment of hypertension

Mode of Action

• Selective alpha-1-adrenoceptors blocker
• Relaxation of smooth muscle in blood vessels and the prostate
• Reduction of pressure on the urethra
• Induction of apoptosis of prostate cells

Dosage and Administration

• Initial dose of 1 mg at bedtime for all patients
• Subsequent dose increase to achieve desired response
• Recommended dose range of 5-10 mg for BPH
• Recommended dose range of 2-10 mg for hypertension

Interaction

• Potential interaction with ACE inhibitors or diuretics
• No interaction with analgesics/anti-inflammatory, cardiac glycosides, hypoglycemic, antiarrhythmic, anxiolytics/sedatives, antibacterial, hormones/steroids, and drugs used for gout

Contraindications

• Hypersensitivity to Terazosin or its analogues

Side Effects

• Postural hypotension
• Dizziness
• Lack of energy
• Peripheral edema
• Urinary frequency
• Priapism

Pregnancy and Lactation

• Not recommended during pregnancy unless benefits justify potential risks
• Caution advised during breastfeeding

Precautions and Warnings

• Risk of hypotension with first dose
• Caution with other antihypertensive agents
• Avoid hazardous tasks when starting therapy
• Dosage reduction and retitration may be necessary when combining with diuretic or other antihypertensive agent

Overdose Effects

• Acute hypotension
• Cardiovascular support is of first importance
• Restoration of blood pressure and normalization of heart rate
• Expansion of plasma volume and noradrenergic vasopressor may be needed
