Indications

  • Single and mixed infestations of hookworm, roundworm, threadworm, whipworm, strongyloides, tapeworm, opisthorchi, hydatid

Description

  • Benzimidazole anthelmintic
  • Active against most nematodes and some cestodes
  • Vermicidal, ovicidal, and larvicidal activity
  • Inhibitory effect on tubulin polymerization
  • Loss of cytoplasmic microtubules

Pharmacology

  • Broad spectrum anthelmintic
  • Block glucose uptake in susceptible helminths
  • Depletion of energy level in helminths
  • Immobilization and parasite death
  • Extensively metabolized, probably in the liver
  • Poorly absorbed from the gastrointestinal tract
  • Rapid first-pass metabolism
  • Principal metabolite with anthelmintic activity

Dosage & Administration

  • 400 mg single dose for various infections
  • 400 mg daily for 3 days in strongyloidiasis or taeniasis
  • 200 mg single dose for children of 1-2 years
  • Not recommended for children under 1 year
  • Dosing for hydatid disease and giardiasis

Interaction

  • No reported pharmacodynamic or pharmacokinetic interaction

Contraindications

  • Not normally used in neonates
  • Dose reduction in children weighing less than 10 kg
  • Not recommended during pregnancy or lactation
  • No need for dose alteration in renal, hepatic, or cardiac failure
