Indications
- Treats HR-positive, HER2-negative advanced breast cancer
- Can be used with aromatase inhibitor or fulvestrant
Pharmacology
- Inhibitor of Cyclin-dependent kinases 4 and 6
- Reduces cellular proliferation of ER-positive breast cancer
- Blocks progression of the cell cycle
Dosage & Administration
- Recommended dose of 125 mg capsule once daily for 21 days followed by 7 days off
- Should be taken with food
- Dose modifications for adverse reactions
Interaction
- Metabolized by CYP3A and Sulfotransferase enzymes
- Affected by CYP3A inhibitors and inducers
- Alters plasma concentrations of certain drugs
Side Effects
- Decreased WBC, Neutrophils, Platelets
- Increased AST, ALT
- Common side effects include infections, fatigue, nausea

- Neutropenia as the most frequent adverse reaction
