Medicine Details

Indications

  • Relief from chronic heartburn symptoms and other symptoms associated with GERD
  • Healing of erosive esophagitis
  • Maintenance of healing of erosive esophagitis
  • Eradication of Helicobacter pylori infection in patients with duodenal ulcer disease
  • Zollinger-Ellison Syndrome
  • Acid-related Dyspepsia
  • Duodenal & Gastric ulcer

Pharmacology

  • Proton pump inhibitor
  • Suppresses gastric acid secretion
  • Has specific inhibition of the H+/K+-ATPase in the gastric parietal cell
  • Absorption occurs approximately 1.5 hours after oral administration
  • Drug metabolism by cytochrome P450 enzyme system

Dosage

  • Different dosages for various conditions
  • Oral dosage form for healing of erosive esophagitis, symptomatic GERD, etc.
  • IV Injection or IV infusion for certain conditions
  • Pediatric dosage based on body weight

Administration

  • Tablet or capsule should be swallowed whole and taken one hour before a meal
  • Oral suspension in a small glass containing water, should be stirred well
  • IV Injection prepared with 0.9% Sodium Chloride for intravenous injection
  • IV injection must be administered over at least 3 minutes

Interaction

  • Metabolized in the liver by CYP2C19 and CYP3A4
  • No clinically relevant interactions with certain drugs
  • May interfere with drugs where gastric pH impacts bioavailability
  • Co-administration effects with clarithromycin

Contraindications

  • Contraindicated in patients with known hypersensitivity to any of the formulation

  • Instructions for patients regarding capsule consumption and use of antacids

Use in Special Populations

  • Safety and effectiveness in pediatric patients have not been established
  • No overall differences in safety and efficacy observed between the elderly and younger individuals

Overdose Effects

  • Lethal effects observed in rats at high doses
  • No specific antidote known
  • Treatment should be symptomatic and supportive

  • Store at a temperature not exceeding 30°C
  • Protect from light and moisture
  • Keep out of reach of children
