Description

Indications
- Relief from chronic heartburn symptoms
- Healing of erosive esophagitis
- Maintenance of healing of erosive esophagitis
- Eradication of Helicobacter pylori infection
- Treatment of Zollinger-Ellison Syndrome
- Symptomatic relief for Acid-related Dyspepsia
- Treatment of Duodenal & Gastric ulcer
Pharmacology
- Proton pump inhibitor
- Suppresses gastric acid secretion
- Better acid control with single optical isomer
- Specific inhibition of the H+/K+-ATPase
- Enteric-coated pellet formulation
- Absorption peaks at approximately 1.5 hours
- Extensively metabolized in the liver
Dosage
- Oral dosage form
- Healing of Erosive Esophagitis: 20 mg or 40 mg Once Daily for 4-8 Weeks
- Maintenance of Healing of Erosive Esophagitis: 20 mg Once Daily
- Symptomatic GERD: 20 mg Once Daily for 4 Weeks
- Helicobacter Pylori eradication: Esomeprazole 40 mg Once Daily for 10 days
- Zollinger-Ellison Syndrome: 20-80 mg once daily
- Acid-related Dyspepsia: 20-40 mg once daily for 2-4 weeks
- Duodenal ulcer: 20 mg once daily for 2-4 weeks
- IV Injection/infusion also available for certain conditions
Administration
- Tablets or capsules to be swallowed whole before a meal
- Oral suspension to be taken with water or administered through a tube
- IV injection solution for intravenous administration
- Specific instructions for nasogastric or gastric tube administration
Interaction
- Metabolized in the liver by CYP2C19 and CYP3A4
- No clinically relevant interactions with specific CYP enzymes
- May interfere with absorption of drugs impacted by gastric pH
Contraindications
- Contraindicated in patients with known hypersensitivity to any of the formulation
Side effects
- Common adverse events: headache, diarrhea, nausea, flatulence, abdominal pain, constipation, dry mouth
Pregnancy lactation
- No adequate studies in pregnant women
- No teratogenic effects in animal studies
- Excretion in milk not studied
- Breastfeeding should be discontinued if essential
Precautions warnings
- Response to therapy does not preclude presence of gastric malignancy
- Specific instructions for patients with difficulty swallowing capsules
- May be used with antacids
Special populations
- Safety and effectiveness not established in paediatric patients
- No overall differences in safety and efficacy observed between elderly and younger individuals
Overdose effects
- Single oral dose at 510 mg/kg lethal to rats
- Major signs of toxicity include reduced motor activity, respiratory frequency changes, tremor, ataxia, and convulsions
- No specific antidote known
- Treatment should be symptomatic and supportive

- Store at temperature not exceeding 30°C
- Protect from light and moisture
- Keep out of reach of children
