Indications
- Relief from chronic heartburn symptoms and symptoms associated with GERD

Pharmacology
- Proton pump inhibitor
- Specific inhibition of the H+/K+-ATPase in the gastric parietal cell
- Absorption peak plasma levels occur at approximately 1.5 hours
- Metabolized in the liver by the cytochrome P450 (CYP) enzyme system
- Plasma elimination half-life of approximately 1–1.5 hours
Dosage
- For healing of Erosive Esophagitis: 20 mg or 40 mg Once Daily for 4-8 Weeks
- 20 mg Once Daily for maintenance of healing of erosive esophagitis
- 20 mg Once Daily for Symptomatic GERD for 4 Weeks
- Triple Therapy for Helicobacter Pylori eradication
- 20-80 mg once daily for Zollinger-Ellison Syndrome
- 20-40 mg once daily for Acid-related Dyspepsia for 2-4 weeks
- 20 mg once daily for 2-4 weeks for Duodenal ulcer and 20-40 mg once daily for 4-8 weeks for Gastric ulcer
- IV Administration for adults - 20 mg or 40 mg once daily
- IV Administration for pediatric patients based on body weight
- 80 mg IV infusion for risk reduction of rebleeding of Gastric or Duodenal Ulcers following therapeutic endoscopy in adults
Administration
- Tablet or capsule should be swallowed whole and taken one hour before a meal
- Oral Suspension administration instructions
- IV Injection administration and storage instructions
Interaction
- Extensively metabolized in the liver by CYP2C19 and CYP3A4
- No clinically relevant interactions with certain drugs
- Potential interference with the absorption of drugs where gastric pH is important
Contraindications
- Contraindicated in patients with known hypersensitivity to any of the formulation

Pregnancy & Lactation
- No well-controlled studies in pregnant women
- No studies on excretion in milk

Use in Special Populations
- Safety and effectiveness in pediatric patients not established
- No overall differences in safety and efficacy between elderly and younger individuals
- No dosage adjustment recommended for patients with mild to moderate hepatic insufficiency
- No alterations in pharmacokinetics in patients with renal impairment
Overdose Effects
- Signs of acute toxicity in rats
- No reports of overdose
- No specific antidote known
- Management should be symptomatic and supportive
