Indications
- Management of moderate to severe acute post-operative pain
- Relief of pain, inflammation, and light sensitivity after ocular surgery
- Decreasing eye inflammation or redness caused by seasonal allergies
Description
- Belongs to pyrrolo-pyrrole group of nonsteroidal anti-inflammatory drug (NSAID)
- Chemical name: 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid, compound with 2 - amino-2-(hydroxymethyl)-1,3-propanediol (1:1)
- Inhibits synthesis of prostaglandins as a peripherally acting analgesic
- Biologically active form: S-form
- Linear pharmacokinetic property
- Highly protein bound
- Metabolized in the liver
- Metabolites and some unchanged drugs excreted in urine
Pharmacology
- Potent analgesic of the non-steroidal anti-inflammatory drugs (NSAIDs)
- Inhibits the cyclooxygenase enzyme system
- Minimal anti-inflammatory effect at analgesic dose
Dosage & Administration
- Tablet: Recommended dose is 10 mg every 4-6 hours for short-term use (up to 7 days)
- Tablet: Not recommended for chronic use or doses exceeding 40 mg/day
- Injection: Used for moderately severe, acute pain management, usually postoperative
- Injection: Administered as single or multiple doses
- Injection: IV bolus given over no less than 15 seconds, IM administration given slowly and deeply into muscle
- Eye Drops: Adults - 1 drop in each eye 4 times daily
Interaction
- Other NSAIDs or aspirin increases side effects of Etorac
- Anti-coagulants enhance anti-coagulant effect
- Beta Blocker reduces anti-hypertensive effect
- ACE Inhibitors increase risk of renal impairment
- Methotrexate enhances toxicity of methotrexate
Contraindications
- Hypersensitivity to Ketorolac or other NSAIDs
- Use not recommended in children under 16 years
- Contraindicated as prophylactic analgesic before surgery

Precautions & Warnings
- Exercise caution in patients over 65 years
- Caution in patients with active or suspected peptic ulcer, gastrointestinal bleeding, asthma, or liver dysfunction
- For Eye Drops: Discontinue use with evidence of corneal epithelial breakdown, monitor closely for corneal health, do not use while wearing contact lenses
