Composition
- 375 mg tablet: Amoxicillin 250 mg + Clavulanic Acid 125 mg
- 625 mg tablet: Amoxicillin 500 mg + Clavulanic Acid 125 mg
- 1 gm tablet: Amoxicillin 875 mg + Clavulanic Acid 125 mg
- Powder for Suspension: Amoxicillin 125 mg + Clavulanic Acid 31.25 mg
- Powder for Suspension (Forte): Amoxicillin 400 mg + Clavulanic Acid 57.5 mg
- 1.2 gm Injection: Amoxicillin Sodium 1 gm + Clavulanate Potassium 200 mg
- 0.6 gm injection: Amoxicillin Sodium 500 mg + Clavulanate Potassium 100 mg
Pharmacology
- Antibacterial combination of Amoxicillin and Clavulanic Acid
- Broad spectrum of bactericidal activity
- Inactivation of beta-lactamase enzymes
- Optimized absorption at the start of a meal
- Closely matched pharmacokinetics of the two components
Dosage
- Different dosages for adults and children
- Various dosage forms: tablet, suspension, forte suspension, IV injection
- Adjusted dosage for more severe infections
- Oral form can be taken without regard to meals, but enhanced absorption at the start of a meal
Administration
- Oral dosage form can be taken without regard to meals
- IV injection not suitable for intramuscular or subcutaneous administration
- Reconstituted vial to be used within 20 minutes, any unused material should be discarded
Interaction
- Prolongation of bleeding time and prothrombin time reported
- Possible reduction of efficacy of oral contraceptives
- Concomitant use of allopurinol can increase allergic skin reactions
- No data on concomitant use of Co-amoxiclav and allopurinol
Contraindications
- History of Penicillin hypersensitivity
- Possible cross-sensitivity with other beta-lactam antibiotics
- Contraindicated for patients with previous Co-amoxiclav or Penicillin-associated cholestatic jaundice
Side Effects
- Uncommon and mild side effects
- Gastrointestinal side effects can be reduced by taking with meals
- Rare cases of hepatitis, cholestatic jaundice, and skin reactions reported
- Rare cases of angioedema and anaphylaxis reported
Pregnancy & Lactation
- Animal studies show no teratogenic effect
- Limited use in human pregnancy with no untoward effects reported
- Not recommended in pregnancy unless considered essential
- Trace quantities detected in breast milk during lactation
Precautions & Warnings
- Careful use in patients on anticoagulation therapy or with severe hepatic dysfunction
- Dosage adjustment for patients with moderate or severe renal impairment
- Adequate fluid intake and urinary output maintenance during high dose administration
Use in Special Populations
- Dose adjustment for patients with renal impairment and hepatic impairment
- Reduced dosage for children with renal or hepatic impairment
Overdose Effects
- Unlikely occurrence of overdose problems
- Possible gastrointestinal symptoms and electrolyte imbalances
- Potential removal from circulation by haemodialysis
Therapeutic Class

Reconstitution
- IV injection reconstitution with Water for Injection BP
- Not suitable for mixing with certain solutions
- Reconstituted solution can be injected into infusion fluids over a specified period
Storage Conditions
- Store below 25°C, protected from light and moisture
- Reconstituted suspension to be kept in the refrigerator and used within 7 days
- Reconstituted vial must be used within 20 minutes
