Indications

• Short-term treatment of bacterial infections
• Upper respiratory tract infections
• Lower respiratory tract infections
• Genito-urinary tract infections
• Skin and soft tissue infections
• Bone and joint infections
• Other infections

Composition

• 375 mg tablet
• 625 mg tablet
• 1 gm tablet
• Powder for Suspension
• Powder for Suspension (Forte)
• 1.2 gm Injection
• 0.6 gm injection

Pharmacology

• Broad spectrum of bactericidal activity
• Susceptible to degradation by (3-lactamases
• Inactivation of beta-lactamase enzymes
• Optimized absorption at the start of a meal
• Low levels of serum binding

Dosage

• Usual adult dose: 625 mg Tablet every 12 hours
• For more severe infections: 1 gm Tablet every 12 hours
• Children dosage based on age and weight
• IV injection dosage for adults and children

Administration

• Oral dosage form can be taken without regard to meals
• Enhanced absorption when taken at the start of a meal
• IV injection suitable for intravenous administration

Interaction

• Prolongation of bleeding time and prothrombin time reported
• Reduced efficacy of oral contraceptives
• Potential interaction with allopurinol

Contraindications

• History of Penicillin hypersensitivity
• Cross-sensitivity with other beta-lactam antibiotics

Side Effects

• Uncommon and mainly of a mild and transitory nature
• Gastrointestinal side effects may be reduced by taking with meals
• Rare cases of hepatitis, cholestatic jaundice, and skin reactions reported

Pregnancy & Lactation

• Animal studies show no teratogenic effect
• Limited human use in pregnancy with no untoward effect
• Trace quantities detected in breast milk

Precautions & Warnings

• Use with care in patients on anticoagulation therapy
• Dosage adjustment for patients with hepatic or renal impairment
• Maintain adequate fluid intake and urinary output during high dose administration

Use in Special Populations

• Dosage adjustment for patients with renal impairment
• Use with caution in patients with hepatic impairment

Overdose Effects

• Unlikely to occur
• Gastrointestinal symptoms and fluid imbalance may be evident
• Can be removed by haemodialysis

Reconstitution

• IV injection reconstitution with Water for Injection BP
• Not to be mixed with certain solutions or fluids
• Can be injected into infusion fluids over a period of 3-4 minutes

Storage Conditions

• Store below 25°C, protected from light and moisture
• Reconstituted suspension should be kept in the refrigerator and used within 7 days
• Reconstituted vial must be used within 20 minutes
