Dosage & Administration

  • 200mg and 400mg tablet

Indications

  • Treatment of Bronchial asthma
  • Bronchospasm

Pharmacology

  • Doxophylline - novel bronchodilator with a dioxolane group in position 7
  • Selective inhibitor of phosphodiesterase 4
  • Relaxes bronchial smooth muscle
  • Decreased affinities toward adenosine A1 and A2 receptors
  • Inhibits platelet activating factor (PAF) and leukotriene production

Interaction

  • Not to be administered with other xanthine preparations
  • Limit consumption of caffeine-containing beverages and food
  • Exercise caution when administered with sympathomimetic drugs and specific medications such as erythromycin, TAO, lincomycin, clindamycin, allopurinol, cimetidine, influenza vaccine, propanolol, phenytoin, and other anticonvulsants
  • Dosage adjustment may be necessary in certain cases

Side Effects & Overdose

  • Possible side effects include nausea, vomiting, epigastric pain, headaches, insomnia, tachycardia, and more
  • Overdose may lead to severe cardiac arrhythmias and tonic-clonic seizures
  • No specific antidote available for overdose

Contraindications

  • Hypersensitivity to the drug or other xanthine derivatives
  • Acute myocardial infarction
  • Hypotension
  • During lactation

Pregnancy & Lactation

  • Use during pregnancy to be carefully evaluated based on risk-benefit ratio
  • Contraindicated during lactation
  • No interference with pre and postnatal growth in animal tests

Precautions & Warnings

  • Factors affecting hepatic clearance may influence drug plasma levels
  • Caution in patients with cardiac disease, hypertension, elderly, severe hypoxemia, hyperthyroidism, chronic corpulmonale, congestive heart failure, liver disease, peptic ulcer, and renal impairment
  • Special caution in congestive heart failure patients due to slower drug clearance
  • No risk of addiction or dependence

Overdose Effects

  • Severe cardiac arrhythmias and tonic-clonic seizures may occur
  • Discontinuation of treatment may be necessary if signs of toxicity appear
  • Symptomatic treatment of cardiovascular collapse in case of overdose
