Description
- Used to treat bronchial asthma, bronchospasm, chronic obstructive pulmonary disease (COPD), pulmonary disease with spastic bronchial component
- Novel bronchodilator with decreased affinities toward adenosine A1 and A2 receptors
- Selectively inhibits phosphodiesterase 4 to relax bronchial smooth muscle
- Dosage for elderly: 200 mg tablet two or three times daily
- Dosage for adults: 400 mg tablet two or three times daily
- Dosage for children: varies based on age and body weight
- Should not be administered with other xanthine preparations
- Interaction caution with ephedrine or other sympathomimetic drugs
- Contraindicated in individuals with known hypersensitivity to the drug or other xanthine derivatives, acute myocardial infarction, hypotension, and during lactation
- Side effects may include nausea, vomiting, epigastric pain, headache, irritability, insomnia, tachycardia, extrasystoles, tachypnea, hyperglycemia, albuminuria, severe cardiac arrhythmias, and tonic-clonic seizure
- Caution advised during pregnancy and lactation
- Does not affect the ability to drive or use machines
- Precautions for administering to patients with various conditions such as cardiac disease, hypertension, hypoxemia, hyperthyroidism, chronic corpulmonale, congestive heart failure, liver disease, peptic ulcer, and renal impairment
- In case of overdose, severe cardiac arrhythmias and tonic-clonic seizure may occur

Indications

Pharmacology
- Selectively inhibits phosphodiesterase 4 to relax bronchial smooth muscle
- Differs from Theophylline due to the presence of a dioxolane group in position 7
- Decreased affinities toward adenosine A1 and A2 receptors
- Inhibits platelet activating factor (PAF) and generation of leukotriene production
Dosage and administration
- Elderly: 200 mg tablet two or three times daily
- Adults: 400 mg tablet two or three times daily
- Children: dosage varies based on age and body weight
- If required daily dose is 400 mg, Doxophylline SR tablet can be taken once daily or as prescribed by the physician
Interaction
- Should not be administered with other xanthine preparations
- Caution advised with ephedrine or other sympathomimetic drugs
- Concurrent administration of certain drugs may reduce hepatic clearance of xanthine derivatives
- Certain factors such as age, congestive cardiac decompensation, chronic obstructive pulmonary disease, severe liver disease, concomitant infections, and the administration of specific drugs may reduce the hepatic clearance of xanthine derivatives
