Indications
- Seasonal and Perennial Allergic Rhinitis
- Chronic Idiopathic Urticaria
Pharmacology
- Ebastine is a long-acting and selective H1-histamine receptor antagonist.
- Inhibition of peripheral receptors remains at a constant level after repeated administration.
- Rapid absorption and extensive first-pass metabolism following oral administration.
- Ebastine is almost totally converted to the pharmacologically active acid metabolite, carebastine.
Dosage & Administration
- Tablet – Adults (> 12 years):
- 10 mg (one tablet) once daily
- Tablet – Children (6-12 years):
- 5 mg (half tablet) once daily
- Syrup – Children (2-5 years):
- 2.5 ml once daily (up to 5 ml in severe cases)
- Syrup – Children (6-12 years):
- 5 ml once daily (up to 10 ml in severe cases)
- Administration:
- May be taken with or without food
Interaction
- Combination with ketoconazole or erythromycin increases plasma level of Ebasten and prolongs QTc interval.
- No interaction with the pharmacokinetics of theophylline, warfarin, cimetidine, diazepam, or alcohol.
- Enhanced sedation effect of alcohol and diazepam.
Contraindications
- Known hypersensitivity to Ebastine or any of its ingredients
Side Effects
- Common: headache, dry mouth, drowsiness
- Less common: abdominal pain, dyspepsia, nausea, insomnia
Overdose Effects
- No clinically meaningful signs or symptoms observed up to 100 mg given once daily.
- No specific antidote for Ebasten.
- In case of accidental overdoses, gastric lavage, monitoring of vital functions including ECG, and symptomatic treatment should be carried out.
