Indications
- Bronchial asthma

Pharmacology
- Novel bronchodilator
- Selective inhibition of phosphodiesterase 4
- Relaxes bronchial smooth muscle
- Decreased affinities towards adenosine A1 and A2 receptors
- Inhibition of platelet activating factor (PAF)
- Inhibition of leukotriene production
Dosage and Administration
- Elderly dosage: 200 mg tablet two or three times daily
- Adults dosage: 400 mg tablet two or three times daily
- Children dosage: >12 years - 10 ml syrup or 200 mg tablet two or three times daily, 6-12 years - 6-9 mg/kg body weight two times daily
- Doxophylline SR tablet for 400 mg daily dose
Interaction
- Avoid admin with other xanthine preparations
- Limit caffeine-containing beverages and food
- Caution with ephedrine or other sympathomimetic drugs
- Concurrent administration may affect hepatic clearance
- Increase in dosage with concurrent phenytoin, anticonvulsants, or cigarette smoking
Contraindications
- Known hypersensitivity to the drug or other xanthine derivatives
- Acute myocardial infarction
- Hypotension
- Lactation

- Insomnia
- Tachycardia
- Extrasystoles

- Hyperglycemia
- Albuminuria
- Severe cardiac arrhythmias

Pregnancy & Lactation
- Use during pregnancy should be evaluated based on risk-benefit ratio
- Contraindicated during lactation
- No interference with pre and postnatal growth in animal tests
Precautions & Warnings
- Age, congestive heart failure, chronic obstructive pulmonary disease, hepatic diseases may reduce clearance
- Caution in patients with cardiac disease, hypertension, hypoxemia, hyperthyroidism, liver disease, renal impairment
- Slow clearance in congestive heart failure patients
- No risk of addiction or dependence
Overdose Effects
- Severe cardiac arrhythmias
- Tonic-clonic seizures
- Discontinue treatment and resume at lower doses after subsiding of symptoms
