Description
- Ruvastin is indicated for treating primary hyperlipidemia and mixed dyslipidemia, hypertriglyceredemia, primary dysbetalipoproteinemia, homozygous familial hypercholesterolemia, atherosclerosis progression, pediatric patients with familial hypercholesterolemia, and risk reduction of myocardial infarction, stroke, and arterial revascularization procedures.
DosageRange
- 5-20 mg/day for pediatric patients (10-17 years of age)
- 40 mg once daily for very high cholesterol and high risk of MI or stroke
- 20 mg once daily for homozygous familial hypercholesterolemia
- 5 mg once daily initiation for Asian patients
- Limited to 5 mg once daily with cyclosporine
- Limited to 10 mg once daily with Lopinavir/Ritonavir or Atazanavir/Ritonavir
- Limited to 10 mg once daily with concomitant lipid-lowering therapy
- Not exceed 10 mg once daily for patients with severe renal impairment
Pharmacology
Selective and competitive inhibitor of HMG-CoA reductase, increasing hepatic LDL receptors and inhibiting hepatic synthesis of VLDL and LDL particles.
Interaction
- Combination with Cyclosporine increases exposure
- Combination with Gemfibrosil should be avoided
- Combination with Lopinavir/Ritonavir or atazanavir/ritonavir increases exposure
- Prolongs international normalized ratio (INR) with coumarin anticoagulants
- Concomitant use of fibrates and niacin products may increase the risk of skeletal muscle effects
Contraindications
- Known hypersensitivity to product components
- Liver disease with unexplained persistent elevations in hepatic transaminase levels
- Pregnant women and women who may become pregnant
- Nursing mothers

- Administered with caution to women of childbearing age
- Discontinue if patient becomes pregnant
- Not known whether excreted in human milk
PrecautionsAndWarnings
- Prescribed with caution in patients with predisposing factors for myopathy
- Discontinue if markedly elevated creatinine kinase levels occur
- Temporarily withheld in any patient with acute, serious condition suggestive of myopathy
- Liver enzyme tests should be performed before and following the initiation of therapy
OverdoseEffects
No specific treatment, symptomatic treatment and supportive measures instituted, hemodialysis does not significantly enhance clearance
