Indications

Pharmacology
- Proton pump inhibitor
- Specific inhibition of the H+/K+-ATPase

- Enteric-coated pellet formulation of esomeprazole magnesium
- Extensively metabolized in the liver by the cytochrome P450 enzyme system
Dosage
- Oral dosage form for various conditions
- IV Injection and IV infusion for specific patient groups
Administration
- Tablet or capsule should be swallowed whole and taken one hour before a meal
- Oral Suspension administration and IV Injection instructions
Interactions
- Metabolism by CYP2C19 and CYP3A4
- No clinically relevant interactions with specific drugs
- Potential interference with CYP2C19 and gastric pH-dependent drugs
Contraindications
- Known hypersensitivity to any of the formulation

Use in Special Populations

- No overall differences in safety and efficacy observed in the elderly
- No dosage adjustment recommended for patients with mild to moderate hepatic insufficiency
- Pharmacokinetics not expected to be altered in renal impairment
Overdose Effects
- Signs of acute toxicity in animal studies
- No specific antidote known
- Expected protein binding limits removal by dialysis
- Treatment should be symptomatic and supportive
