Indications
- Bronchial asthma
- Bronchospasm
- Chronic obstructive pulmonary disease (COPD)
- Pulmonary disease with spastic bronchial component
Pharmacology
- Novel bronchodilator
- Inhibits phosphodiesterase 4
- Relaxes bronchial smooth muscle
- Selective inhibition of adenosine A1 and A2 receptors
- Inhibits platelet activating factor (PAF)
- Inhibits leukotriene production
Dosage & Administration
- Elderly: 200 mg tablet two or three times daily
- Adults: 400 mg tablet two or three times daily
- Children over 12 years: 10 ml syrup or 200 mg tablet two or three times daily
- Children 6-12 years: 6-9 mg/kg body weight two times daily
- Doxophylline SR tablet for required daily dose of 400 mg
Interaction
- Not to be administered with other xanthine preparations
- Limit consumption of caffeine-containing beverages and food
- Caution with ephedrine or other sympathomimetic drugs
- Caution with concurrent administration of erythromycin, TAO, lincomycin, clindamycin, allopurinol, cimetidine, influenza vaccine, propanolol, phenytoin, and other anticonvulsants
- Cigarette smoking may increase clearance of xanthine derivatives
Contraindications
- Known hypersensitivity to the drug or other xanthine derivatives
- Acute myocardial infarction
- Hypotension
- During lactation

Pregnancy & Lactation
- Animal tests show no interference with pre and postnatal growth
- Clinical evidence lacking for effects during pregnancy
- Contraindicated during lactation
- No effect on ability to drive or use machines
Precautions & Warnings
- Factors reducing hepatic clearance may increase plasma levels
- Caution in patients with cardiac disease, hypertension, the elderly, severe hypoxemia, hyperthyroidism, chronic corpulmonale, congestive heart failure, liver disease, peptic ulcer, and renal impairment
- Monitoring of blood levels recommended
- Caution in patients with congestive heart failure
Overdose Effects
- Severe cardiac arrhythmias
- Tonic-clonic seizure
- Symptomatic treatment of cardiovascular collapse in case of overdose
