Indications
- Treatment of invasive aspergillosis
- Treatment of candidemia in nonneutropenic patients
- Treatment of disseminated candidiasis in skin, abdomen, kidney, bladder wall and wounds
- Treatment of esophageal candidiasis
- Treatment of serious infections caused by Scedosporium apiospermum and Fusarium Species
- Treatment for patients intolerant of, or refractory to other therapy
Pharmacology
- Triazole antifungal medication
- Binds and inhibits ergosterol synthesis
- Inhibits CYP450-dependent 14-alpha sterol demethylase
- Results in depletion of ergosterol in fungal cell membrane
Dosage & Administration
- Oral administration
- Requires at least one hour gap before or after a meal
- Different dosing for patients above and below 40kg body weight
- Loading and maintenance dose regimen for different conditions
- Injection for invasive aspergillosis, candidemia, Scedosporiosis and Fusariosis
Interaction
- Interaction with CYP3A4, CYP2C9, and CYP2C19 inhibitors and inducers
- Potential increase in concentrations and activity of drugs that are CYP3A4, CYP2C9, and CYP2C19 substrates
- Co-administration with Phenytoin or Efavirenz increases maintenance oral and intravenous dosage of Vorizol
Contraindications
- Known hypersensitivity to Voriconazole
- Co-administration with specific drugs like terfenadine, astemizole, cisapride, pimozide, quinidine, rifampin, carbamazepine, long-acting barbiturates, efavirenz, ritonavir, rifabutin, ergot alkaloids, and St. John's Wort

Pregnancy & Lactation
- No adequate and well-controlled studies in pregnant women

- Reported cases of squamous cell carcinoma of the skin with long-term Vorizol treatment

Reconstitution
- Instructions for reconstituting suspension
- Shake the bottle well before adding water
- Add specific amount of boiled and cooled water to the bottle
