Indications

  • Bronchial asthma

Pharmacology

  • Doxophylline is a novel bronchodilator
  • Inhibits phosphodiesterase 4
  • Relaxes bronchial smooth muscle
  • Selectively inhibits phosphodiesterase 4
  • Decreased affinities toward adenosine A1 and A2 receptors
  • Inhibits platelet activating factor (PAF) and generation of leukotriene production

Dosage & Administration

  • Dosage for Elderly: 200 mg tablet two or three times daily
  • Dosage for Adults: 400 mg tablet two or three times daily
  • Dosage for Children (>12 years of age): 10 ml syrup or 200 mg tablet two or three times daily
  • Dosage for Children (6-12 years of age): 6-9 mg/kg body weight two times daily
  • Doxophylline SR tablet for required daily dose of 400 mg

Interaction

  • Avoid administration with other xanthine preparations
  • Limit consumption of beverages and food containing caffeine
  • Use caution when combining with ephedrine or other sympathomimetic drugs
  • Concurrent administration with erythromycin, TAO, lincomycin, clindamycin, allopurinol, cimetidine, influenza vaccine, and propanolol
  • Phenytoin, other anticonvulsants, and cigarette smoking may affect clearance
  • Possible dosage adjustment in case of interaction

  • Known hypersensitivity to the drug or other xanthine derivatives
  • Acute myocardial infarction
  • Hypotension
  • Lactation
