Indications
- Bronchial asthma
- Bronchospasm
- Chronic obstructive pulmonary disease (COPD)
- Pulmonary disease with spastic bronchial component
Pharmacology
- Doxophylline is a novel bronchodilator
- Selectively inhibits phosphodiesterase 4
- Relaxes bronchial smooth muscle
- Inhibits platelet activating factor (PAF) production
- Decreased affinities toward adenosine A1 and A2 receptors
Dosage & Administration
- 200 mg tablet for elderly two or three times daily
- 400 mg tablet for adults two or three times daily
- 10 ml syrup or 200 mg tablet for children over 12 years old
- 6-9 mg/kg body weight for 6-12 years old
- Doxophylline SR tablet for daily 400 mg dose
Interaction
- Avoid administering with other xanthine preparations
- Limit consumption of caffeine-containing beverages and food
- Exercise caution with ephedrine or other sympathomimetic drugs
- Concurrent administration may reduce hepatic clearance
- Some drugs may increase clearance of xanthine derivatives
Contraindications
- Known hypersensitivity to the drug or other xanthine derivatives
- Acute myocardial infarction
- Hypotension
- During lactation

Pregnancy & Lactation
- Animal tests show no interference with pre and postnatal growth

- Contraindicated during lactation

- Factors may reduce hepatic clearance of xanthine derivatives
- Caution in administering to patients with various conditions
- Monitoring blood levels recommended in certain cases
- No risk of addiction or dependence
Overdose Effects
- Severe cardiac arrhythmias
- Tonic-clonic seizure
- Discontinuation may be required
- Symptomatic treatment for cardiovascular collapse
