Indications

  • Bronchial asthma
  • Bronchospasm
  • Chronic obstructive pulmonary disease (COPD)
  • Pulmonary disease with spastic bronchial component

Pharmacology

  • Novel bronchodilator
  • Selectively inhibits phosphodiesterase 4
  • Inhibition of platelet activating factor (PAF)
  • Decreased affinities toward adenosine A1 and A2 receptors

Dosage & Administration

  • Elderly: 200 mg tablet two or three times daily
  • Adults: 400 mg tablet two or three times daily
  • Children: >12 years of age: 10 ml syrup or 200 mg tablet two or three times daily
  • Children: 6-12 years of age: 6-9 mg/kg body weight two times daily
  • Doxophylline SR tablet to be taken once daily if required daily dose is 400 mg

Interaction

  • Not to be administered with other xanthine preparations
  • Limit consumption of beverages and food containing caffeine
  • Exercise caution with other sympathomimetic drugs
  • Concurrent administration of drugs that may reduce hepatic clearance

Contraindications

  • Known hypersensitivity to the drug or other xanthine derivatives
  • Acute myocardial infarction
  • Hypotension
  • During lactation

  • Tachycardia
  • Insomnia
  • Tachypnea
  • Hyperglycemia
  • Albuminuria
  • Severe cardiac arrhythmias
  • Tonic-clonic seizure

Pregnancy & Lactation

  • Use during pregnancy should be evaluated carefully based on risk-benefit ratio
  • Contraindicated during lactation
  • Does not affect ability to drive or use machines

Precautions & Warnings

  • Factors affecting hepatic clearance
  • Monitoring of blood levels in specific cases
  • Caution in patients with cardiac disease, hypertension, hypoxemia, etc.
  • Use with caution in congestive heart failure patients

Overdose Effects

  • Severe cardiac arrhythmias
  • Tonic-clonic seizure
  • Discontinuation of treatment may be necessary
  • Symptomatic treatment of cardiovascular collapse
