Indications

  • Short-term management of moderate to severe acute post-operative pain
  • Relief of pain, inflammation, and light sensitivity after ocular surgery
  • Decreasing eye inflammation or redness caused by seasonal allergies

Description

  • Belongs to the pyrrolo-pyrrole group of nonsteroidal anti-inflammatory drug (NSAID)
  • Chemical composition: 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid, compound with 2 - amino-2-(hydroxymethyl)-1,3-propanediol (1:1)
  • Inhibits synthesis of prostaglandins
  • Biological activity associated with the S-form
  • Linear pharmacokinetic property, highly protein bound and largely metabolized in the liver
  • Metabolites and some unchanged drugs excreted in the urine

Pharmacology

  • Potent analgesic of the non-steroidal anti-inflammatory drugs (NSAIDs)
  • Acts by inhibiting the cyclooxygenase enzyme system
  • Minimal anti-inflammatory effect at its analgesic dose

Dosage & Administration

  • Tablet dosage: 10 mg every 4-6 hours, short-term use only (up to 7 days)
  • Injection dosage: Single or multiple doses for the management of moderately severe, acute pain
  • Eye Drops dosage: 1 drop in each eye, 4 times daily

Interaction

  • Other NSAIDs or aspirin: Increase the side effects of Rotalac
  • Anti-coagulants: Enhance anti-coagulant effect
  • Beta Blocker: Reduce the anti-hypertensive effect
  • ACE Inhibitors: Increase the risk of renal impairment
  • Methotrexate: Enhance the toxicity of methotrexate

Contraindications

  • Hypersensitivity to the drug or other NSAIDs
  • Not recommended for use in children under 16 years of age
  • Contraindicated as a prophylactic analgesic before surgery

Precautions & Warnings

  • Caution in patients over the age of 65 years
  • Caution in patients with active or suspected peptic ulcer, gastrointestinal bleeding, asthma, and liver dysfunction
  • Discontinue use of topical NSAIDs and monitor corneal health in patients with evidence of corneal epithelial breakdown
