Indications
- First-line treatment for metastatic non-small cell lung cancer (NSCLC)
- Treatment of patients with metastatic EGFR T790M mutation-positive NSCLC
Pharmacology
- Kinase inhibitor of the epidermal growth factor receptor (EGFR)
- Exhibits anti-tumor activity against NSCLC lines harboring EGFR-mutations
Dosage & Administration
- Recommended dose of 80 mg tablet once a day
- Can be taken with or without food
- Instructions for administration via nasogastric tube
Interaction
- Avoid concomitant administration with strong CYP3A inhibitors
- Avoid concomitant administration with strong CYP3A inducers
Side Effects
- Common adverse reactions include diarrhea, rash, dry skin, nail toxicity, stomatitis, fatigue, decreased appetite
Pregnancy & Lactation
- Can cause fetal harm when administered to a pregnant woman
- Advise females of reproductive potential to use effective contraception
- May impair fertility in females and males of reproductive potential
Precautions & Warnings
- Risk of interstitial lung disease (ILD)/pneumonitis
- Monitor for QTc interval prolongation
- Assess left ventricular ejection fraction (LVEF) before treatment
- Can cause embryo-fetal toxicity
Use in Special Populations
- No clinically significant differences in pharmacokinetics based on age, sex, ethnicity, body weight, baseline albumin, smoking status
- No established safety and effectiveness in pediatric patients
- Higher incidence of Grade 3 and 4 adverse reactions in patients 65 years or older
- No recommended dose adjustments for patients with renal impairment
