Indications

  • Osteoarthritis

Pharmacology

  • Immediate release Esomeprazole Magnesium layer
  • Enteric-coated Naproxen core
  • Proton pump inhibitor
  • Inhibition of prostaglandin synthesis

Dosage

  • Lowest effective dose
  • Tablet dosage for different conditions and age groups
  • Consideration for different treatment if lower Esomeprazole dose is more appropriate

Administration

  • Take at least 30 minutes before meals
  • Do not split, chew, crush or dissolve tablet

Interaction

  • Reduced antihypertensive effect with concomitant use of NSAIDs and ACE inhibitors, diuretics & beta-blockers
  • Increased bleeding risk with concomitant use of warfarin
  • Interference with drug absorption due to Esomeprazole inhibiting gastric acid secretion
  • Effects of high-fat food on absorption of Naproxen & Esomeprazole

Contraindications

  • Hypersensitivity to tablet components or substituted benzimidazoles
  • History of allergic-type reactions after taking aspirin or other NSAIDs
  • Use during peri-operative period in coronary artery bypass graft (CABG) surgery

Side Effects

  • Decreased occurrence of gastric ulcers and NSAID associated upper gastrointestinal adverse events
  • Risk of arterial thrombotic events with long-term high dose use
  • Gastrointestinal adverse events
  • Oedema, hypertension, cardiac failure
  • Adverse effects on fertility

Pregnancy & Lactation

  • Caution during first and second trimester
  • Contraindicated during third trimester
  • Excretion in human milk
  • Effect on fertility
