Indications

  • Relief from chronic heartburn symptoms

Pharmacology

  • Proton pump inhibitor
  • Suppresses gastric acid secretion
  • Absorption in 1.5 hours
  • Enteric-coated pellet formulation
  • Extensive metabolism in the liver
  • Excretion primarily in urine and faeces
  • Combination therapy with antimicrobials

Dosage

  • 20 mg or 40 mg oral dosage for erosive esophagitis
  • 20 mg oral dosage for symptomatic GERD
  • Triple Therapy for Helicobacter pylori eradication
  • 20-80 mg dosage for Zollinger-Ellison Syndrome
  • 20-40 mg dosage for acid-related dyspepsia
  • 20 mg dosage for 2-4 weeks for duodenal ulcer

Administration

  • Oral tablet or capsule should be swallowed whole
  • Oral suspension to be taken with water
  • IV injection or IV infusion for specific conditions

Interactions

  • Extensive metabolism by CYP2C19 and CYP3A4
  • No clinically relevant interactions with certain drugs
  • Potential interference with CYP2C19 and drugs affected by gastric pH
  • No change in pharmacokinetic profile with certain co-administered drugs

Contraindications

  • Known hypersensitivity to the formulation

  • Lethal overdose in rats at 103 times the human dose
  • Symptomatic and supportive treatment recommended
