Description
- Oral capsule for the treatment of heartburn, GERD, erosive esophagitis, H. pylori infection, Zollinger-Ellison Syndrome, Dyspepsia, Duodenal & Gastric ulcers
Indications
- Relieves chronic heartburn symptoms
- Heals erosive esophagitis
- Maintains healing of erosive esophagitis
- Eradicates Helicobacter pylori infection
- Treats Zollinger-Ellison Syndrome
- Manages Acid related Dyspepsia
- Heals Duodenal & Gastric ulcers
Pharmacology
- Supresses gastric acid secretion
- Proton pump inhibitor
- Enters systemic circulation within 1.5 hours after administration
- Metabolized in the liver
- Extensively bound to plasma proteins
- Excreted as inactive metabolites in the urine and feces
Dosage
- 20 mg or 40 mg once daily for 4-8 weeks for healing erosive esophagitis
- 20 mg once daily for maintenance of healing of erosive esophagitis
- 20 mg once daily for symptomatic GERD
- Esomeprazole 40 mg Once Daily for 10 days in combination therapy for H.pylori eradication
- 20-80 mg once daily for Zollinger-Ellison Syndrome
- 20-40 mg once daily for acid-related Dyspepsia
- 20 mg once daily for 2-4 weeks for Duodenal ulcer, 20-40 mg once daily for 4-8 weeks for Gastric ulcer
- Dosage adjustments for pediatric and adult patients based on body weight
- 80 mg for adults for risk reduction of rebleeding of Gastric or Duodenal Ulcers following therapeutic endoscopy
Administration
- Oral capsule should be swallowed whole and taken 1 hour before meals
- Oral suspension should be taken with water and stirred well
- IV injection or infusion for specific conditions and patient demographics
Interaction
- Extensively metabolized in the liver by CYP2C19 and CYP3A4
- No clinically relevant interactions with certain drugs metabolized by specific CYP enzymes
- Potential interference with absorption of drugs affected by gastric pH
- No significant interactions with specific drugs like phenytoin, warfarin, clarithromycin or amoxicillin
Contraindications
- Contraindicated in patients with known hypersensitivity to any of the formulation
Side effects
- Frequently occurring adverse events include headache, diarrhea, nausea, flatulence, abdominal pain, constipation and dry mouth
Pregnancy and lactation
- No adequate and well-controlled studies in pregnant women
- No teratogenic effects observed in animal studies
- Excretion in milk not studied
- Breast-feeding should be discontinued if use is considered essential
Precautions and warnings
- Symptomatic response to therapy does not preclude the presence of gastric malignancy
- Specific instructions for patients regarding capsule administration and use of antacids
- Paediatric and geriatric use, hepatic and renal insufficiency considerations

- Lethal to rats at high doses
